Seventy-one pounds of average weight loss and a clean Phase 3 sweep — the succession plan filed before the throne is even contested.
Retatrutide is the succession plan for the best drug franchise in pharma: a triple agonist (GLP-1, GIP and glucagon) that has now swept its pivotal program. TRIUMPH-4 paired 28.7% weight loss with a 76% reduction in knee-osteoarthritis pain; TRIUMPH-1 delivered the program's largest average loss — up to 71.2 pounds1; and TRIUMPH-2 and -3 met every primary endpoint in July 2026, extending the sweep to diabetic and cardiovascular populations, with regulatory filings for obesity, sleep apnea and osteoarthritis submitted on the results2.
If approved on schedule, Lilly will own both the most effective injectable ever measured and the most convenient pill — the top and bottom of the market, before Mounjaro's patents even approach their cliff. Seven further Phase 3 readouts arrive by year-end, each a chance to extend labels or stumble publicly. The honest risks are tolerability at the doses that produce record numbers, and pricing power against payers who already groan at incretin budgets. Retatrutide's late-stage trials are part of this bill. R&D growing faster than revenue for a year would mean the next generation is costing more to reach market.
